Characterization of Chemical Compounds and Antioxidant Activity of Centaurea Solstitialis ssp. Schouwii (DC.) Q. Et S. (Asteraceae)

Abstract:


Background: A detailed study was performed on the antioxidant activity, the total phenolic and total flavonoid contents of the derived extracts (chloroform, ethyl acetate and n-butanol) of the 70% ethanol extract of the aerial parts of Centaurea solstitialis growing in Algeria with the aim of selecting the most active extracts for phytochemical investigations.

Methods: The antioxidant abilities were assessed using 1,1-diphenyl-2-picrylhydrazyl radical (DPPH•) scavenging essay, and cupric ion reducing antioxidant capacity (CUPRAC). Butylhydroxyanisole (BHA), Butylhydroxytoluene (BHT) and α –Tocopherol were used as positive controls. The total phenolic and total flavonoid contents of the extracts were also determined.

Results: The ethyl acetate (EtOAc) extract exhibited the highest activities followed by the n-butanol (n-BuOH) extract wile this last extract had the highest phenolic and flavonoid contents followed by EtOAc extract. These data allowed guiding the fractionation of EtOAc and n-BuOH extracts by chromatographic methods leading to a new and a known sesquiterpene lactones together with 3 known flavonoid glycosides.

Conclusion: A new guainolide named 3-(4-hydroxybenzoyl)-cynaratriol was isolated and identified together with 4 known secondary metabolites. The structures were established by spectral analyses mainly high resolution electrospray ionisation mass spectrometry (HR-ESIMS) and one- and two-dimensional nuclear magnetic resonance spectroscopy experiments.
